After Simon agreed to take part, the boys then signed up some of the biggest stars in telly who have each appeared on screen alongside the music mogul - and have had beef with him at some point.
They include Louis Walsh, Caroline Flack and Olly Murs from The X Factor and former girlfriend Sinitta. And from Britain’s Got Talent, judges Amanda Holden and Alesha Dixon, ITV2 host Stephen Mulhern, former panellist Piers Morgan and previous winners Diversity and canine star Pudsey and handler Ashley Butler.
Standing at the top of a decadent staircase, the 55-year-old is seen looking down on his famous friends and colleagues as they sing Happy Birthday To You to him.
But, channelling his inner Mr Nasty, he stops the sing-a-long with an unimpressed swipe of his hands as he huffs: 'That is so obvious... Second song?'
As the likes of Ant and Dec, Louis Walsh and his Britain's Got Talent co-stars Amanda Holden and Alesha Dixon look befuddled at his bizarre request, the lights in the mansion dim.
A gunshot is then heard, a flash of light seen and screams throughout the room before Simon is seen in pain and tumbling down the curved staircase.
Simon's right-hand woman Sinitta - clad in her famous X Factor leaves costume - comes bolting forward in a panic.
'Someone shot Simon Cowell!' she screams, before Christopher Biggins pipes up, 'We know dear! We all saw it.'
The funny thread - which will be hosted by former Pussycat Doll star Ashley Roberts - will play out in typical style over the course of the popular weekend TV show, and see detectives Emilia Fox and Kevin Whately try to piece together the clues to find the murderer.
Who Shot Simon Cowell – mirroring the famous Dynasty plot Who Shot JR – will play out over five weeks before the culprit is finally revealed. Simon and his celebrity pals filmed the sketch in a north London mansion on Monday.
An inside source told The Sun “There are a lot of stars who have had run-ins with Cowell over the years – but who would have a grudge big enough to want to shoot him?
“That is the tantalising premise of the sketch. “It was Ant and Dec’s idea and they put it to Cowell who thought it was great fun. ”Everyone really got into the spirit of it during filming on Monday night.
